Se caracteriza por ser hidrof\u00edlica y por su forma activa homodim\u00e9rica. Al contrario de la mayor\u00eda de las bacteriocinas descritas, Lcn972 no forma poros en la membrana citoplasm\u00e1tica sino que inhibe parcialmente la incorporaci\u00f3n de precursores de la pared celular y bloquea la formaci\u00f3n del septo de divisi\u00f3n. Nuestros resultados recientes han demostrado que Lcn972 es la primera bacteriocina no modificada descrita que interacciona espec\u00edficamente con el l\u00edpido II, mol\u00e9cula universal y esencial precursora de la s\u00edntesis de pared en bacterias. Sin embargo, se desconoce la naturaleza de dicha interacci\u00f3n y la base molecular del restringido espectro de actividad de Lcn972. En este proyecto, abordaremos ambos aspectos apoy\u00e1ndonos en el conocimiento generado por nuestro grupo sobre Lcn972 (maquinaria biosint\u00e9tica, producci\u00f3n a gran escala) y sus caracter\u00edsticas (ausencia de modificaciones post-traduccionales y su modo de acci\u00f3n espec\u00edfico) de modo que Lcn972 pueda convertirse en una mol\u00e9cula modelo para el desarrollo de nuevos antibi\u00f3ticos. Nos planteamos resolver la estructura tridimensional de Lcn972 en soluci\u00f3n por RMN y complementar el estudio mediante la caracterizaci\u00f3n funcional de mutantes de esta prote\u00edna. Estas dos estrategias estructural y funcional se integrar\u00e1n para entender la base molecular de la interacci\u00f3n Lcn972-l\u00edpido II y dise\u00f1ar estrategias para mejorar su estabilidad y actividad. En segundo lugar, se investigar\u00e1 sobre la entidad molecular que determina la especificidad de acci\u00f3n de Lcn972.
